ScienceDaily (Aug. 28, 2011) ? Most cells rely on structural tethers to position chromosomes in preparation for cell division. Not so oocytes. Instead, a powerful intracellular stream pushes chromosomes far-off the center in preparation for the highly asymmetric cell division that completes oocyte maturation upon fertilization of the egg, report researchers at the Stowers Institute for Medical Research.
Their findings illustrate how oocytes repurposed a dynamic cellular mechanism capable of generating considerable intracellular forces and widely used by migrating cells to propel them forward, to set the stage for asymmetric cell division -- the kind of cell divisions that generate two different daughter cells. It might also lead to improvements in the selection criteria used to choose the most promising oocytes for in-vitro fertilization.
As a mammalian egg develops, it undergoes two highly asymmetric cell divisions, known as meiosis I and II. During each of these divisions, the cytoplasm divides unequally, giving rise to a large egg and two polar bodies that are much smaller than the developing oocyte. To achieve this uneven distribution of the cytoplasm, the meiotic spindle -- the structure that separates the chromosomes into daughter cells -- has to be positioned close to the so-called cortical cap, the region where the polar body will form.
"Conventional thinking predicted some sort of physical tether that moors the meiotic spindle at the cortical cap," says Rong Li, Ph.D., Stowers investigator and senior author of the study published in the August 28, 2011, advance online edition of Nature Cell Biology. "It came quite as a surprise that, instead, a continuous intracellular flow pushes the spindle into the correct position and keeps it there."
Earlier studies had ruled out microtubules, which help position the spindle during mitotic cell divisions, as potential tethers while older studies had hinted at actin as a possible candidate. Actin, one of the most abundant protein in animal cells, forms dynamic filament networks that play a crucial role in many cellular processes, including cell migration, intracellular transport.
To find out whether and how actin might play a role in the position of the meiotic spindle, postdoctoral researcher and first author Kexi Yi, Ph.D., incubated mouse oocytes with several known inhibitors of the actin cytoskeleton. "Within minutes of applying CK-666, a brand new and very specific inhibitor, the spindle drifted away from the cortical cap towards the center of the oocyte."
CK-666 inhibits the Arp2/3 complex, a major regulator of the actin cytoskeleton that is known to play a role in cell locomotion and membrane trafficking. It binds to existing actin filaments and initiates the growth of new "branch" filaments. Further experiments revealed that myosin-II contractility, better known for producing muscle contractions, pushes the spindle away from the cortical cap when the Arp2/3 complex is inhibited.
Previous work by Li and her team had shown that meiotic chromosomes, when positioned close to the cortex of an oocyte in meiosis II, induces the formation of a cortical actin cap by propagating the regulatory signal from the Ran protein. When Yi tested the effect of intercepting the Ran signal, they found that Ran also regulates Arp2/3 localization and by extension, spindle position.
Yi then turned to high-resolution time-lapse confocal microscopy and spatiotemporal correlation spectroscopy (STICS), in collaboration with Stowers imaging experts, Jay Unruh, Ph.D., and Brian Slaughter, Ph.D., both co-authors on the paper, to track the dynamics of the cytoplasmic actin network in oocytes labeled with a live F-actin probe.
"STICS analysis showed that the actin flow originates at the cortical cap and continues down along both sides of the lateral cortex before it converges near the center of the oocyte and reverses direction toward the spindle," says Yi. When he treated the oocytes with jasplakinolide, an actin filament-stabilizing drug, actin flow in the cells' interior almost immediately ceased.
"The actin flow drives cytoplasmic streaming away from the cortical cap region along the cell periphery. When it arrives at the opposite pole of the oocyte, it circulates back in a pattern similar to that of the actin flow toward the spindle," says Li. A theoretical analysis by physicist and co-author Boris Rubinstein, Ph.D., a research advisor at the Stowers Institute, found that the observed cytoplasmic streaming generates pressure on the spindle and pushes it towards the cortex.
In many vertebrate species including mammals, oocytes may arrest in meiosis II for hours or even days awaiting fertilization. "During this time the asymmetric spindle position must be stably maintained," explains Li. "Maintaining spindle position under an active force could prevent slow and random drift of spindle position or orientation if the meiotic arrest is prolonged."
Loss of asymmetric positioning of the meiosis II spindle is a known cause of impaired reproductive potential in aging females and spindle position is used as a clinical index to evaluate the quality of oocytes arrested in meiosis II for in-vitro fertilization.

MIAMI ? A weakened but still dangerous Hurricane Irene shut down New York and menaced other cities more accustomed to snowstorms than tropical storms as it steamed up the East Coast on Saturday, unloading a foot of rain on North Carolina and Virginia and knocking out power to 2 million homes and businesses. At least eight people were killed.
New York emptied its streets and subways and waited with an eerie quiet. Washington braced for the onslaught, too, as did Philadelphia, the New Jersey shore and the Boston metropolitan area. Packing wind gusts of 115 mph, the hurricane had an enormous wingspan ? 500 miles ? and threatened a swath of the nation inhabited by 65 million people.
The hurricane stirred up seven-foot waves. At New York Harbor, even with the eye of the storm several hours away, there was a 3.5-foot storm surge early Sunday.
Across the Northeast, drenched by rain this summer, the ground is already saturated, raising the risk of flooding as well as the danger of trees falling onto homes and power lines.
Irene made its official landfall just after first light near Cape Lookout, N.C., at the southern end of the Outer Banks, the ribbon of land that bows out into the Atlantic Ocean. While it was too early to assess the full extent of damage, shorefront hotels and houses were lashed with waves, two piers were destroyed and at least one hospital was forced to run on generator power.
"Things are banging against the house," Leon Reasor said as he rode out the storm in the town of Buxton, N.C. "I just hate hurricanes."
Eastern North Carolina got 10 to 14 inches of rain, according to the National Weather Service. Virginia's Hampton Roads area was drenched with at least nine inches, with 16 reported in some spots.
By late Saturday night, the storm had sustained winds of 80 mph, down from 100 mph on Friday. That made it a Category 1, the least threatening on a 1-to-5 scale, and barely stronger than a tropical storm.
Nevertheless, it was still considered highly dangerous, capable of causing ruinous flooding across much of the East Coast with a combination of storm surge, high tides and 6 to 12 inches of rain.
There were no hurricane-force winds over land Sunday morning; they were limited to a relatively small area over the Atlantic Ocean as the eye of the storm traveled over water after touching down in North Carolina. But tropical-storm-force winds still covered a wide area of land, and Irene was still expected to be a hurricane when its eye made landfall again in the New York area around noon Sunday.
Irene was moving north-northeast at 17 mph, slightly faster than it had been Saturday, and was centered about 195 miles south-southwest of New York City. A typical hurricane would be moving much faster, 25 to 30 mph, said senior hurricane specialist Stacy Stewart of the National Hurricane Center.
Pennsylvania Gov. Tom Corbett warned that the state will not necessarily be out of danger once the storm has passed: "The rivers may not crest until Tuesday or Wednesday. This isn't just a 24-hour event."
As of Saturday evening, Irene was hugging the U.S. coastline on a path that could scrape every state along the Eastern Seaboard. Ed Rappaport, deputy director of the National Hurricane Center in Florida, said it would be a "low-end hurricane, high-end tropical storm" by the time it crossed the New York City area late Sunday morning.
The storm is so large that areas far from Irene's center are going to be feeling strong winds and getting large amounts of rain, he said.
"It is a big, windy, rainy event," he said.
The deaths blamed on Irene included two children, an 11-year-old boy in Virginia killed when a tree crashed through his roof and a North Carolina child who died in a crash at an intersection where traffic lights were out. Four other people were killed by falling trees or tree limbs ? two in separate Virginia incidents, one in North Carolina and one in Maryland. A surfer and another beachgoer in Florida were killed in heavy waves.
Power outages were concentrated in Virginia and North Carolina.
Irene was the first hurricane to make landfall in the continental United States since 2008, and came almost six years to the day after Katrina ravaged New Orleans on Aug. 29, 2005. Experts guessed that no other hurricane in American history had threatened as many people.
North Carolina Gov. Beverly Perdue said Irene inflicted significant damage along her state's coast, but that some areas were unreachable because of high water or downed power lines. "Folks are cut off in parts of North Carolina, and obviously we're not going to get anybody to do an assessment until it's safe," she said.
At least 2.3 million people were under orders to move to somewhere safer, though it was unclear how many obeyed or, in some cases, how they could.
Defense Secretary Leon Panetta told 6,500 troops from all branches of the military to get ready to pitch in on relief work, and President Barack Obama visited the Federal Emergency Management Agency's command center in Washington and offered moral support.
"It's going to be a long 72 hours," he said, "and obviously a lot of families are going to be affected."
In New York, authorities undertook the herculean job of bringing the city to a halt. The subway began shutting down at noon, the first time the system was closed because of a natural disaster.
On Wall Street, sandbags were placed around subway grates near the East River because of fear of flooding. Tarps were spread over other grates. Construction stopped throughout the city, and workers at the site of the World Trade Center dismantled a crane and secured equipment.
The city was far quieter than on an average Saturday. In some of the busiest parts of Manhattan, it was possible to cross a major avenue without looking, and the waters of New York Harbor, which might normally be churning from boat traffic, were quiet. About 370,000 people living in low-lying areas of the city, mostly in Lower Manhattan, Brooklyn and Queens, were under orders to clear out.
"The time to leave is right now," Mayor Michael Bloomberg said at an outdoor news conference at Coney Island, his shirt soaked from rain.
The New York area's major airports ? LaGuardia, Kennedy and Newark ? waved in their last arriving flights around noon. The Giants and Jets postponed their preseason NFL game, the Mets postponed two baseball games, and Broadway theaters were dark.
New York has seen only a few hurricanes in the past 200 years. The Northeast is much more used to snowstorms ? including the blizzard last December, when Bloomberg was criticized for a slow response.
Airlines said 9,000 flights were canceled, including 3,000 on Saturday. The number of passengers affected could easily be millions because so many flights make connections on the East Coast.
Greyhound suspended bus service between Richmond, Va., and Boston. Amtrak canceled trains in the Northeast for Sunday.
In Philadelphia, Mayor Michael Nutter declared a state of emergency, the first for the city since 1986, when racial tensions were running high. "We are trying to save lives and don't have time for silliness," he said.
The storm arrived in Washington just days after an earthquake damaged some of the capital's most famous structures, including the Washington Monument. Irene could test Washington's ability to protect its national treasures and its poor.
In New Jersey, the Oyster Creek nuclear plant, just a few miles from the coast, shut down as a precaution as Irene closed in. And Boston's transit authority said all bus, subway and commuter rail service would be suspended all day Sunday.

Two of my favorite New Mexico rivers to fish are the San Juan River and Cimarron River. This story is about the Cimarron. The Cimarron River is in Northeast New Mexico and emanates out of Eagle Nest Lake. Located on the eastern edge of the Sangre de Cristo Mountains, this dry fly gem flows eastward through Cimarron Canyon State Park along US Hwy 64. After flowing into several other rivers this water ultimately reaches the Mississippi River. New Mexico Game and Fish estimates nearly 4,000 catchable sized wild browns per mile, this is unquestionably the healthiest wild brown trout fishery in the state and one of the healthiest in the southern Rocky Mountains. Most of the wild browns are 10-14 inches with the occasional big boy. This fishery is not well known but can still get a little crowded close to the roads during mid summer.
I can easily get to the Cimarron within a one-day hard drive from my Fly Shop near Tyler, Texas and be fishing late that afternoon or early the next morning. The high mountain scenery is beautiful and the fishing is great.
My story begins in the middle of June 2002. During June, the stonefly hatch is in full swing on the Cimarron and big browns will eagerly attack a size 14 or 16 Stimulator with regularity. This is my favorite time of year to fish this river. This is dry fly fishing at its best.
Unfortunately, if you remember, this was during the time wild fires were engulfing many parts of New Mexico, Colorado and Arizona. The Cimarron Canyon State Park was surrounded by a couple of such fires.
I arrived at the Pine Ridge Hotel, only about a mile from quality fishing water, late Monday evening. This is a quaint little four-room hotel nestled near the entrance of the state park. After checking in, the hotel owner told me that the park was closed and had been since Friday. Because of the fire dangers, it was closed to everything including camping, hiking, picnicking and YES? fishing. Every park pull off was barricaded in bright yellow. It seems like every other tree had a Park Closed sign nailed to it. I couldn?t believe it. I had driven 12 hours strait to spend some personal time dry fly fishing and? wouldn?t you know it? No Fishing. I was sick. If you have ever wanted to see a big man cry this would have been a great opportunity.
There were no other places near by that I could fish unless I had a guide to fish some private waters that were near by. I called my good friend, Doc Thompson, of High Country Anglers. Doc is the best fly fishing guide for the small streams of Northeast New Mexico including some private areas of the Cimarron. He was booked. No chance for me to fish any private water.
I was considering cutting my losses, packing up my gear and heading home with my tail tucked between my legs when I saw the park ranger turn into the motel parking lot. I eagerly approached him, explained my predicament and pleaded with him to be merciful on this misguided Texan. He indicated that there was little he could do. He was ordered by higher-ups to close the park. The area was very dry and the fear of the forest fire spreading was at an all time high. He finally gave me a miniscule window of hope when he said that if the wind didn?t pick up or change directions during the night he would ask if he could open the park the next day. No guarantees. There was only a small chance of things falling my way but I decided to wait it out and see.
The next morning, bight and early, I walked to the hotel owners desk with my fingers and toes crossed. ?Great news? the owner said, ?you can fish.? The park ranger called the hotel and told the owner to go tell Tex he can fish. The ranger opened the park to fishing only. No one could camp, hike or picnic. Fishing only. Best of all, he left all of the barricades and Park Closed signs up. My head began to buzz with this scenario. Could it be true? Is it possible that any where in this country there is a stream that is full of big browns, full of fish eager to slam dry flies, has had no fishing for five days, has Park Closed signs on every other tree and has bright yellow barricades at every pull off? Is this really happening? And most of all? is this really happening to me? YES? it was!!!
I almost broke a leg and a fly rod getting to river so quick. I was the only person fishing this 12-mile stretch of quality trout water. The ONLY person. The river has had no fishing pressure for five days. None. Notta. Absolutely zero. What a magnificent opportunity. I hit the water at 8:00 am. Knowing that the stonefly hatch would began at a bout 9:00 am, I tied on a size 16 yellow Stimulator with a Copper John dropper on my 2wt rod. For the next hour, the catch was 50/50 on the dry and nymph. When the hatch started, I removed the nymph in order concentrate on the dry. My stimulator was repeatedly demolished by hungry trout. The hatch started around 9:00 am and eventually tapered off around 2:00 pm as the day warmed. During that time, I landed more than 40 nice brown on a dry. Most of them were in the 10 to 12 inch range. Some a little smaller and some a little bigger. The whole time, I saw no other person on the river. I was in total fly fishing bliss. Later that afternoon, as the temperature began to drop, they started hitting hard again and did so until just before dark. I lost count of the total number of fish that I caught that day. After a while you just quit counting. The fishing was phenomenal and the solitude was even better.
During the day, when I would take brake at my parked Jeep for a shot of Joe or something, people would drive buy giving me the stare of disgust. I could see their lips mumble something like, ?that idiot Texan? can?t he read the signs?? I was in no way interested in setting them strait. I wanted the entire state on New Mexico as well as the whole wide world to think the park was completely closed and may never open again.
That evening, back at my room, the harsh realities of that summer were vivid. I sat on my front porch and watched the helicopters and other large aircraft drop loads of water on the forest fires that were scattered along the mountainside. Huge walls of smoke draped the horizon. I couldn?t help but become sorrowed because of the destruction that was unfolding before my eyes. When it was too dark to see, I retired to the confines of my room.
Later that night, as I lay packed tight in my cozy little bed, I found it difficult to sleep. As my mind drifted from the fires outside and danced with thoughts of my day of fishing, I giggled as I felt myself swelling with contentment. My angling experience kept running through my mind. I couldn?t help but wonder what the next day would bring. Would my fly fishing solitude come to an end? Guess what? it didn?t.
I hit the park early the next morning to be faced with the very same scenario. The signs and barricades were still in place and no one was in sight. Using the same flies and techniques I enjoyed equal success as I had the day before. It wasn?t until late that afternoon did I see the only other car in the park. The word was finally out. By then, it was OK. I enjoyed a few hours of good fishing the next morning before heading back to Tyler.
Is there a moral here? I don?t know. It was defiantly a fishing trip that I will never forget. In retrospect, however, it seems really unfortunate that my memorable fishing trip was at the expense of so much. The southwest lost an awful lot of good forest and helpless wildlife during that horrible time. Homes were gone and lively hoods were destroyed. I suppose the saying remains true, ?someone?s good fortune is usually at someone else?s expense.?

The tension between kids and teens over who is or should be in charge is related to issue of power, control, experience and personal development.
As teens develop both physically and emotionally and as their awareness of self changes, it can be tempting for teens to compare themselves with others as part of the natural desire to feel accepted by their peers. The brain undergoes many changes during the teen?s years. Teens know what?s best for kids and kids know what?s best for them. Teens have experience and kids want experience.
Teens want to take some time and think it over, and kids want to do it yesterday. Teens want to talk and kids want to act. Teens think about consequences and kids think about now. Teens think they have to be in charge, and kids want a chance to be in charge. Teens can ?hang in there? longer than kids, who get bored easily and quickly and want a change of task or job. Teens think that it will be too expensive and that kids don?t understand it. Teens really don?t respect kids, but expect kids to respect them.
They won?t be teenagers forever, only adults whose brains are rewiring in decision making, impulse control and judgment making connects all of them. Certainly this could account for much of the irrational decisions and mood swings. The world depends on that. The world depends on us!

Lessons from 2001 anthrax case help pinpoint source of disease
FLAGSTAFF, Ariz. Aug. 23, 2011 Employing technology that reads the entire DNA code, researchers led by the Translational Genomics Research Institute (TGen) and the Technical University of Denmark (DTU) have pinpointed the source of a cholera outbreak in Haiti that killed more than 6,000 people and sickened 300,000.
Using whole genome sequencing, which spells out the billions of chemical bases in DNA, TGen and DTU provided the strongest evidence yet that peacekeepers from Nepal, where cholera is widespread, brought the disease to Haiti, which already was reeling from a devastating earthquake in January 2010 that killed more than 300,000.
In a study titled "Population genetics of Vibrio Cholerae from Nepal: An identical clone in Nepal and the Haitian outbreak," researchers confirm the source of the cholera, and suggest how to prevent future outbreaks when international aid is rushed to disaster areas.
The study appears Tuesday August 23rd in mBio, a new online-only, open-access journal published by the American Society of Microbiology in partnership with the American Academy of Microbiology.
"The great similarity of Haitian cholera with Nepalese cholera is based upon the highest resolution DNA methods available, and point to a probable source of this devastating disease outbreak," said Dr. Paul Keim, Director of the TGen Pathogen Genomics Division and senior molecular biologist on the study. Dr. Keim assisted the FBI in tracking down the source of the 2001 anthrax letters case, which killed five people. He said similar genetic tracking techniques were used in investigating the Haitian cholera outbreak.
According to Dr. Keim, methods pioneered during the anthrax letter forensic investigation and today's greatly diminished costs for whole genome sequencing make it possible to apply this powerful technology to new and critical public health challenges. Dr. Keim is a Regents Professor at Northern Arizona University (NAU), whose Center for Microbial Genetics and Genomics also contributed to the study.
Dr. Keim praised TGen's collaborators at the National Public Health Laboratory in Nepal, and at the National Food Institute in Denmark, where the study's senior author, Dr. Frank M. Aarestrup, is head of the Antimicrobial Resistance and Molecular Epidemiology Unit.
"This study highlights how rapidly infectious diseases might be transmitted globally through international travel, and how public health officials need to use advanced molecular tools, along with standard epidemiological analyses, to quickly and accurately determine sources of outbreaks," said Dr. Aarestrup, who also is head of both the World Health Organization's Collaborating Centre for Antimicrobial Resistance among Foodborne Pathogens and of the European Union (EU) Reference Laboratory for Antimicrobial Resistance.
Dr. Lance Price, an associate professor at TGen and co-author of the new study, said the investigation into the source of Haitian cholera could help prevent such outbreaks in the future.
"This effort validates the power of advanced molecular tools in investigating outbreaks of this nature," Dr. Price said. "The goal now should be finding ways to prevent such outbreaks, perhaps through screening prior to deployment. This study is not about placing blame, it's about preventing such disasters in the future."
Researchers confirmed the source of the outbreak by comparing the DNA of 24 cholera samples (the bacterium Vibrio cholera) from five different districts in Nepal with 10 samples of cholera from Haiti. All 24 samples from Nepal matched the samples from Haiti. Some of the samples, the report said, "were almost identical."
Dr. Price said the TGen findings makes a very strong case for the source of the cholera, and aligns with a recent report by the U.S. Centers for Disease Control and Prevention, which said "evidence strongly suggests" that the Nepalese soldiers were the source of the outbreak. The CDC study was based on the fact that the first cholera infections in Haiti were downstream from the Nepalese, and occurred shortly after the soldiers' arrival.
"Our studies are complementary and together make a definitive case for a Nepalese origin to the outbreak," Dr. Price said.

Dr. Keim praised TGen's collaborators at the National Public Health Laboratory in Nepal, and at the National Food Institute in Denmark, where the study's senior author, Dr. Frank M. Aarestrup, is head of the Antimicrobial Resistance and Molecular Epidemiology Unit.
"This study highlights how rapidly infectious diseases might be transmitted globally through international travel, and how public health officials need to use advanced molecular tools, along with standard epidemiological analyses, to quickly and accurately determine sources of outbreaks," said Dr. Aarestrup, who also is head of both the World Health Organization's Collaborating Centre for Antimicrobial Resistance among Foodborne Pathogens and of the European Union (EU) Reference Laboratory for Antimicrobial Resistance.
Dr. Lance Price, an associate professor at TGen and co-author of the new study, said the investigation into the source of Haitian cholera could help prevent such outbreaks in the future.
"This effort validates the power of advanced molecular tools in investigating outbreaks of this nature," Dr. Price said. "The goal now should be finding ways to prevent such outbreaks, perhaps through screening prior to deployment. This study is not about placing blame, it's about preventing such disasters in the future."
Researchers confirmed the source of the outbreak by comparing the DNA of 24 cholera samples (the bacterium Vibrio cholera) from five different districts in Nepal with 10 samples of cholera from Haiti. All 24 samples from Nepal matched the samples from Haiti. Some of the samples, the report said, "were almost identical."
Dr. Price said the TGen findings makes a very strong case for the source of the cholera, and aligns with a recent report by the U.S. Centers for Disease Control and Prevention, which said "evidence strongly suggests" that the Nepalese soldiers were the source of the outbreak. The CDC study was based on the fact that the first cholera infections in Haiti were downstream from the Nepalese, and occurred shortly after the soldiers' arrival.
"Our studies are complementary and together make a definitive case for a Nepalese origin to the outbreak," Dr. Price said.
